I think it was Twilight’s reaction that made Celestia laugh and not Discord’s joke.

Celestia nearly blew her ruse there with how she and Twilight were trying to maintain order at the Gala, despite Discord’s attendance there, when she laughed at Discord’s joke at Twilight’s expense.
 
She invited him to liven things up, but had to act like Discord’s presence was more of a burden than a welcoming sight, especially after the Smooze drenches her and everyone else in slime.
 
Once everything was sorted out and amends were made, Celestia dropping the ruse and revealing that she was actually enjoying herself is priceless!!

Celestia was so cute in yesterday’s episode!
