Well, the show does not exactly tell us that Twi was bullied. Just that she had no friends besides Spike. There could have been plently of ponies who could have tried to befriend her and she didn’t took those chances due her study or such stuff.
And sure there was the sequences of “going back to magic kindergarten” but those could be explained by Twi having a fear or just being very imaginative, etc. Not neccessairly bullying.
He has said that he’s actively avoiding the cliche “she was bullied when she was a filly” thing, so… I dunno.
