Possibly, it’s a bit hard to tell. She has serious chemistry, aproaching into romantic subtext with Mako, cultimating in the pictured kiss (but it was never 100% confirmed they became a couple), and when wearing Junketsu shown visible interest in women (but she was brainwashed then).
 
On the other hand, in her dream state, she was to marry a (faceless) guy, but that was a very generalized/archetypal vision, and part of her brainwashing, and she ultimatelly chose Mako over him, when in a wedding gown nevertheless (adding to the above mentioned romantic subtext).
 
So I think it’s accurate say it’s very probable Ryuko is bisexual or homosexual, but it’s somethng the writers left a bit ambigious.
