Well, there’s no way Celestia’s only one thousand years old, so I’m guessing Luna and Celestia only celebrate their birthdays once a millennium.  
Which means that Luna must have missed the last one while exiled. No wonder she looks so happy!

For future reference, Luna, how about just one candle for every hundred years?

I see no blues or sadness here whatsoever. Except Celestia’s worry that Luna will burn something with that cake, but that’s not related to immortality. Neither does the description, as far as Google Translate allows me to understand, imply sadness.
