A Fluttershy with no friends would probably be an immature, pathological liar who acts helpless and feeds on pity. She might return small kindnesses to her ‘friends’ (so long as they continue to view her favorably), but she won’t leave her comfort zone without some kicking and screaming.

CoCo seems a bit like a friendless Fluttershy to me. But unlike the other 3, she’s a good character, so she doesn’t quite fit in. Also she’s generally way better than Fluttershy, which is not the case for these other 3.

I’m gonna go out on a limb, and predict that Weird Al’s character is going to be the “Pinkie Pie without Friends”.

Friendless version of Fluttershy, not sure. Not sure we’d ever see her since she’d be reclusive shut-in who spends her days browsing the Internet.
 
Applejack without friends (or family) would probably be a cut-throat business figure. Probably stopped believing in “hardwork will see you through” and employs dirty tricks and corporate sabotage.
 
Friendless Pinkie… That’s an interesting thought. I could see a scary, desperately lonely pop idol or something who wants to have friends more than anything but constantly drives others away with her clinginess, possibly escalating to her kidnapping other people and keeping them in her basement to be her friends forever.
